破壁法提取平菇多糖的结构分析及其抗氧化活性Structural Characterization and Antioxidant Activity of Pleurotus ostreatus Polysaccharide Extracted by Wall-broken Method

摘要(Abstract):
采用破壁法提取平菇子实体粗多糖,经 Sevag 法、透析、DEAE-52 和 Sephadex G-100 分离纯化后得到 2 个均一组分多糖 POPH-1 和 POPH-2,总糖含量分别为 76.25% 和 81.29%。利用高效凝胶渗透色谱法(HPGPC)、离子色谱法(IC)及红外光谱(FT-IR)分析 POPH-1 和 POPH-2 的结构,并利用体外自由基清除试验对 POPH-1 和 POPH-2 的抗氧化活性进行了比较。结果表明:POPH-1 和 POPH-2 均为吡喃糖,其中POPH-1 分子质量为 5.12×10~5Da,由阿拉伯糖、岩藻糖、甘露糖、半乳糖和葡萄糖,按摩尔比 1.00∶3.66∶46.74∶92.21∶242.52组成;POPH-2分子质量为 3.98×10~5Da,由阿拉伯糖、岩藻糖、果糖、甘露糖、半乳糖和葡萄糖,按摩尔比 1.00∶2.65∶2.97∶4.32∶28.14∶249.62 组成。POPH-2 质量浓度在 0.5~5.0 mg/mL 时,对 DPPH 和 ABTS 自由基的清除率最大,可分别达到 71.6%和92.2%,说明POPH-2具有较强的抗氧化能力。
